If you don’t count again you may not have realised. Essentially they: From a legal advice perspective- I would ask about what the cash handling training is in your company (triple counting, having a second person counting above a certain amount, etc.) and get loss prevention to advise before going back to your manager. If this is a one off, and you’ve not ignored a cash handling policy/training, then it should be an informal conversation and anything else would be a bit heavy handed. If above 2 year service I wouldn’t worry unless it’s a repeated offense, if under 2 years the manager doesn’t need to justify a dismissal so I’d really try to work with them to try and show goodwill.

Also, my store does a really poor job at training. Another sales associate actually accepted euros and old notes. Managers are solely motivated by sales target, and don’t care about anything else. Nobody gets the training they need. I think it may be worth escalating to head office HR or loss prevention! Approach it more as trying to solve a problem (this has happened and I feel the company could benefit from clearer SOPs) rather than an issue with your manager. If they have any common sense you won’t be blamed for it if you’ve bot been trained/properly supervised. Worth speaking to your area/regional manager next time they’re in store! You also need to provide more information because big retail stores such as the one you’ve stated you work for will have a big HR firm and if it truly is going to proceed with a file note/disciplinary, they would’ve need to run it through HR.
